Single-cell proteomics (SCP) requires an extreme balance of acquisition speed, sensitivity, and quantitative fidelity. The Orbitrap Astral Zoom MS addresses these demands through architectural innovations specifically engineered for ultra-low input samples, enabling deep proteome coverage at scales previously unattainable.
This presentation explores how hardware pillars—specifically Bent Trap technology for parallelized ion pre-accumulation and MS2 scan rates of 270 Hz—drive a near 100% duty cycle and a ~30% increase in ion utilization. By leveraging high-speed, narrowwindow Data-Independent Acquisition (nDIA), this workflow provides a robust LabelFree Quantification (LFQ) solution characterized by median CVs of <10% and high intrascan dynamic range. Performance benchmarks demonstrate the system's exceptional sensitivity and depth, consistently identifying and quantifying ~3,700 protein groups at 120 SPD from 50 pg inputs, and scaling to approximately 7,000 proteins from 250 pg HeLa digest. This synergy of hardware and method ensures high data completeness across large cell cohorts, capturing both structural and regulatory proteins with the quantitative rigor required for deep biological discovery.
